How do DSB interact with the nuclear periphery and what are the functional consequences of this position ? The yeast model organism enables to combine tightly controlled cellular system with genetics, high throughput technologies and advanced live microscopy approaches. We will define the spatial and temporal behavior of damaged chromatin at a single cell level along the cell cycle and perform high throughput screens to decipher the molecular pathways controlling DSB position in the nucleus. Both approaches will give hints into the functionnal consequences of DSB localization in the nucleus, functionnal consequences that we will also test directly by assessing DNA repair and checkpoint activation at the cellular level.
